Base Exchange Reaction with Nicotinamide Mononucleotide by NAD Glycohydrolase

Abstract
Exchange of nicotinamide portion of nicotinamide mononucleotide with various pyridine, imidazole, purine and pyrimidine bases by the transglucosidation reaction of NAD glycohydrolase was investigated with a view to establishing that nicotinamide mononucleotide might function as an alternative donor of ribose-5-phosphate group to bases in the formation of ribotide.
The enzyme partially purified from rabbit brain was shown to catalyze the exchange reaction between the nicotinamide moiety of nicotinamide mononucleotide. 10-4-10-3M, and various bases, 10-2-10-4M, in the following percentage (% conversion of free base to the respective ribonucleotide) nicotinamide, 5.10%; 5-amino-4-imidazolecarboxamide, 5%; adenine, 2.5% isonicotinic acid hydrazide, 0.5%; nicotinic acid, 0.2-0.3%; and uracil, 0.1% respectively.
These results suggested that the base exchange reaction with nicotinamide: mononucleotide by NAD glycohydrolase could not be related to the chemical structure of the base, and was not so reactive as the reaction between 5-phosphoribosyl-l pyrophosphate and bases.
